Waterline/bootstripe

If I were to sand my entire hull, bottom and topside. Can I measure down to the current waterline first fore and aft. When I am done sanding. Make a mark then run a chalk line between the two marks? Will this give me an accurate waterline to tape to? Is there a better way? This is a canoe stern boat.

No, do to the curve of the hull.

A way to get a correct waterline is to use water! Get the boat level. Fill a clear hose that can rest on the ground and reach above the waterline with colored water until the water level matches where you want the new waterline to be. Move the hose along the hull on both sides marking the water line.

Tape and paint.

Replace "chalk line" with "laser level" and it will work.
